Firibastat (QGC001), an orally active brain penetrating proagent of EC33, is a first-in-class brain aminopeptidase A (APA) inhibitor (Ki=200 nM). Firibastat selectively and specifically inhibits conversion of brain angiotensin-II into angiotensin-III and decreases blood pressure in hypertensive rats[1][2].

Firibastat (0.1-30 mg/kg；po) 穿过胃肠道和血脑屏障，进入大脑，产生两种EC33活性分子，抑制大脑APA活性，阻断脑血管紧张素III的形成，使高血压大鼠血压降低数小时[2]。
