Beta-glucuronidase is an important lysosomal enzyme involved in the degradation of glucuronate-containing glycosaminoglycan^[1].

活性测定
1.复溶：
将浓度为 2 mg/mL 的酶溶解于冷的 0.15 M 氯化钠中。测定时将酶稀释至浓度范围为 0.25 至 2 mg/mL。
2.步骤
1) 将以下试剂移入一系列编号的试管中：（包括一个试管作为空白）。
0.1 M Sodium acetate, pH 4.5 0.8 mL
0.01 M Phenolphthalein glucuronidate, pH 6.8 0.1 mL
2) 在 37℃ 水浴中孵育 5 分钟以达到温度平衡。每间隔一段时间向每个试管中加入 0.1 mL 酶稀释液。向空白中加入 0.1 mL 水，孵育 30 分钟。
3) 加入 5.0 mL 0.2 M 甘氨酸、0.2 M NaCl、pH 10 终止反应。
4) 从水浴中取出，冷却至室温并读取 A540 与空白对照的数值。
3.计算
绘制标准曲线，1U 为每小时从酚酞葡萄糖醛酸中释放 1 μmoL 酚酞所需的酶量 (mg)。
