凋亡信号调节激酶 1 (ASK1) 是丝裂原活化蛋白激酶 (MAP3K) 家族的成员,可作为 p38 MAPK 和 c-Jun N 末端激酶 (JNK) 激活的上游调节剂。在正常生理条件下,ASK1 通过与还原形式的硫氧还蛋白结合而保持非活性状态。然而,在诱导细胞内产生活性氧物质的病理条件下(例如线粒体或内质网应激),硫氧还蛋白被氧化并与 ASK1 分离,从而促进 ASK1 的激活以及可能导致组织损伤的下游 p38 MAPK 和 JNK 信号。

Apoptosis signal-regulating kinase 1 (ASK1) is a member of the mitogen-activated protein kinase (MAP3K) family which acts as an upstream regulator for the activation of p38 MAPK and c-Jun N-terminal kinase (JNK). Under normal physiological conditions, ASK1 is maintained in an inactive state through binding to the reduced form of thioredoxin. However, during pathological conditions which induce intracellular production of reactive oxygen species (e.g., mitochondrial or endoplasmic reticulum stress), thioredoxin is oxidized and dissociates from ASK1 facilitating activation of ASK1 and downstream p38 MAPK and JNK signaling that can cause tissue injury .
