DAz-2 

DAz-2 is a sulfonic acid probe for living cells, which can directly detect sulfonic acid-modified proteins in living cells and is cell-permeable^[1].

1. 制备 10 μM 蛋白，或 50 μg 细胞裂解液，或 200 μg 蛋白裂解液。
2. 加入 50 或 100 μM H₂O₂ 室温反应 20 min； 然后添加 1 mM Daz-2 或 5% (v/v) DMSO，并在室温下反应 15 min。
3. 用 5 mM DTT 猝灭反应，并通过凝胶过滤除去多余的试剂。
4. 使用 4-12% SDS-PAGE 分离蛋白质，转移至 PVDF 膜，并通过 HRP-Streptavidin WB 检测。
