Human α-thrombin is a multifunctional serine protease that plays a pivotal role in the coagulation pathway. Human α-thrombin cleaves fibrinogen and converts it into fibrin. Human α-thrombin can stimulate platelet activation and stabilize fibrin polymers[1].

α-凝血酶由一条轻链 (A chain) (Mw~6,000) 和一条重链 (B chain) (Mw~31,000) 通过二硫键连接而成。某些情况下，α-凝血酶会发生自溶生成 β-凝血酶和 γ-凝血酶。本品是由匀质化的人凝血酶原经由 Xa 因子，Va 因子和磷脂活化而得，经 SDS-PAGE 检测确保凝血酶原的完全活化，并以 NIH 凝血酶的标准品为参考，活性不低于 2,700 NIH units/mg。

使用说明
凝血酶对不同的融合蛋白切割效率不一样，建议先进行小量酶切试验。比如固定融合蛋白10 μg，凝血酶设置梯度添加量 (如0.1 U, 0.2 U, 0.5 U, 1 U 等)，以及温度梯度 (如4℃，25℃，37℃等) 进行试验。建议摸索好最佳条件后再进行扩大酶切实验。
