meta-iodoHoechst 33258 is a marker dye in Hoechst series. Hoechst is A live nuclear marker dye. Hoechst binds to the grooves in the DNA double strand, which tends to be A/ T-rich DNA strand. Although it binds to all nucleic acids, the A/ T-rich double strand DNA significantly enhances fluorescence intensity Therefore,Hoechst dye can be used for living cell labeling. The fluorescence intensity of Hoechst dye increases with the increase of pH of solution^[1].

通用实验方案
Hoechst工作液的制备
1.1 储存液的配制
将10 mg Hoechst溶解于5 mL去离子水中。
注：建议将储存液避光保存于4°C或-20°C，并避免反复冻融。
1.2 Hoechst工作液的配制
将储存液用无血清细胞培养基或PBS稀释，以获得终浓度为10 μg/mL的Hoechst工作液。
注：请根据实际情况调整Hoechst工作液的浓度。
细胞染色
2.1 悬浮细胞（6孔板）
a. 在4°C条件下以1000 g离心3-5分钟，然后弃上清。用PBS洗涤两次，每次5分钟，细胞密度为1×10⁶/mL。
b. 加入1 mL工作液，室温孵育3-10分钟。
c. 在4°C条件下以400 g离心3-4分钟，然后弃上清。
d. 用PBS洗涤两次，每次5分钟。
e. 用无血清细胞培养基或PBS重悬细胞，通过荧光显微镜或流式细胞术观察。
2.2 附壁细胞
a. 在无菌盖玻片上培养附壁细胞。
b. 从培养基中取出盖玻片并吸去过量的培养基。
c. 加入100 μL工作液，轻轻摇动以完全覆盖细胞，室温孵育3-10分钟。
d. 用培养基洗涤两次，每次5分钟。通过荧光显微镜或流式细胞术观察。
储存条件
4°C，有效期1年。避光保存。
