N-acetylglucosamine-1-P uridyltransferase (AGX1) (EC 2.3.1.157) (GlcNAc1pUT) is a bifunctional acetyltransferase/uridyltransferase. N-acetylglucosamine-1-P uridyltransferase (AGX1) binds GlcNAc-1-P and UTP, and catalyzes an uridyltransfer reaction to synthesize UDP-GlcNAc. N-acetylglucosamine-1-P uridyltransferase (AGX1) is a bifunctional enzyme exclusive to prokaryotes[1].

产品信息
本品利用大肠杆菌重组表达,N-terminal His Tag,分子量 57 kDa,储存缓冲液组分为 20 mM Tris-HCl,200 mM NaCl pH 8.0, 5% 甘油。可用于催化 GlcNAc/GalNAc 形成UDP-GlcNAc/UDP-GalNAc,作为糖基转移酶转移糖基的供体,也可以作为 UDP-GlcNAc、UDP-GalNAc 的合成的工具酶。

使用方法
1.按照反应体系加入底物,5 mM MgCl2,可适当加入缓冲盐如: Tris-HCl,调节 pH 为 7.5-8.5,加入适量酶 (根据反应需求);
2.混匀后,将样品置于 37 ℃ 孵育 30-60 min。在 pH 接近中性 pH 的缓冲中活性最强。推荐反应缓冲是 Tris-HCL (pH 8.0),多数常见的生物缓冲也适用,比如 Tris 或 PBS。

注意事项
体系放大时候,孵育时间或酶量需要根据实际情况调整,可低速震荡混匀,加入 Inorganic pyrophosphatase, Saccharomyces cerevisiae (HY-P2992) 可以增加转化率。
