APAF1 Protein, a pivotal apoptosome component, orchestrates cytochrome c-dependent activation of pro-caspase-9, initiating caspase-3 activation and apoptosis. Forming an apoptosome ring with cytochrome c and dATP, APAF1 interacts with pro-caspase-9, releasing mature caspase-9. Pro-caspase-3 joins the complex, activating caspase-3. Notably, APAF1 isoform 6 is less effective in inducing apoptosis. APAF1 interacts with APIP, NAIP/BIRC1, and CIAO2A, contributing to its multifaceted role in apoptotic regulation.
