IL-1R2 蛋白, Rat (HEK293, His)

IL-1R2 is a competitive endogenous inhibitor of IL-1 signaling. IL-1R2 serves as a decoy receptor and competes with IL-1R1 for IL-1, it can also form a complex with IL-1RAP once it binds IL-1. IL-1R2 is implicated in various IL-1-mediated inflammatory diseases^[1]. IL-1R2 Protein, Rat (HEK293, His) is a recombinant rat IL-1R2 protein with a His tag at the C-terminus and is expressed in HEK293 cells. It consists of 355 amino acids (M1-E355).

IL-1R2 是非信号传导 2 型白细胞介素 1 受体，天然存在于中性粒细胞、B 细胞、单核细胞和巨噬细胞中。大鼠IL-1R2可裂解成两种形式：膜型(14-416个aa)和可溶性(14-?aa)。
大鼠IL-1R2的氨基酸序列相似性较低(约60%）与人 IL-1R2 相比，但与小鼠 IL-1R2 仅略有改变（90% 相似性）。
IL-1R2 含有截短的胞质结构域，并且缺乏 Toll-IL-1 受体 (TIR) 区域，使其无法跨膜信号传导。IL-1R2 是 IL-1 信号传导的内源性抑制剂。功能性 IL-1 信号传导需要 IL-1R1 和 IL-1RAP 的 IL-1 依赖性募集。IL-1R2 作为诱饵受体，可以与 IL-1R1 竞争 IL-1。一旦IL-1R2与IL-1结合，IL-1R2也可以与IL-1RAP形成复合物，从而防止IL-1RAP与IL-1R1异二聚化。通过这两种方式，IL-1R2 阻断 IL-1 信号传导。此外，可溶性 IL-1R2 以高亲和力募集可溶性 IL-1RAP，而不影响对 IL-1RA 的亲和力。

IL-1R2 (8 μg of pCAGGS-IL-1RII-IgGFc; tail vein; once) inhibits IL-1-induced immune responses in Lewis rats^[4].

Measured by its binding ability in a functional ELISA. Immobilized Rat IL-1R2 at 2 μg/mL (100 μL/well) can bind biotinylated Human IL-1 beta. The ED₅₀ for this effect is 19.20 ng/mL.

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH₂O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Stored at -20°C for 2 years. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
