Antiparasitic agent-25 (Compounds 11) is an orally active antiparasitic agent. Antiparasitic agent-25 inhibits both parasite invasion and replication ability and has irreversible action against Toxoplasma gondii, significantly reducing the replication rate of Toxoplasma gondii with an IC50 value of 6.33 μM, and demonstrates low cytotoxicity with a CC50 value of 285 μM[1].

Result: Constrained the replication of the majority of tachyzoites to 0-1 cycles, manifesting a prevalence of vacuoles sized for accommodating only one or two tachyzoites.

Cell Invasion Assay[1]

Incubation Time: 30 min
Result: Significantly cut down the number of invaded parasites, indicating could inhibit the invasion of parasites into host cells.

Antiparasitic agent-25 (50 mg/kg; 口服给药;每天2次,连续5天) 可减少寄生虫数量,统计上显著延长了中位生存期,但不足以完全抑制体内寄生虫的生长,导致持续感染,从而使受感染小鼠有较高的死亡率[1]

Animal Model: Female Kunming mice (eight-nine weeks old) (Inoculated with 1 × 103 tachyzoites of the RH strain via intraperitoneal (ip) injection.)[1]
Dosage: 50 mg/kg (Dissolved in PBS including 25% PEG-400 and 5% DMSO)
Administration: Oral gavage (p.o.); twice daily for 5 days
Result: Reduced parasite numbers, statistically significantly extended the median survival time, but insufficient to completely suppress the growth of parasites in vivo, leading to continued infection and consequently high mortality rates among infected mice.
