Fura-FF pentapotassium 

Fura-FF pentapotassium is a low-affinity fluorescent dye for calcium (Ex/Em: 365/514 nm in the absence of calcium; 339/507 nm in the presence of a high calcium concentration)^[1].

Guidelines^[2] (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
Labeling of Human Sperm Cells:
1. Incubate the cells according to your normal protocol.
2. Load cells with 5 μM Fura-FF in 1 mL Hepes-buffered human sperm (H-HSM) medium at 36 ℃ for 40 min.
3. Wash the sample by centrifugation and scatter cells in disposable centrifuge tubes at 1 10⁷ cells in 3 mL of H-HSM and maintained at 36 ℃.
4. Immediately measure fluorescence recordings.
