CTLA-4 protein inhibits T-cell responses as a critical negative regulator, with a stronger affinity for CD80 and CD86 than CD28. This affinity allows CTLA-4 to effectively suppress T-cell activation and regulate immune responses, maintaining a delicate equilibrium in T-cell-mediated immunity.
