LIX/CXCL5 蛋白, Mouse

CXCL5, also known as neutrophil activating peptide 78 (ENA‐78), is a CXC chemokine containing ELR motif. CXCL5 promotes angiogenesis through interaction with its specific receptor CXCR2. CXCL5 is expressed by many immune cells, such as macrophages, eosinophils, and non-immune cells including mesothelial cells, and fibroblasts.CXCL5/CXCR2 axis not only contributes to the recruitment of neutrophils but also regulates the function of neutrophils in melanoma^[1][2]. LIX/CXCL5 Protein, Mouse is produced in E.coil, and consists of 92 amino acids (A41-Q132).

CXCL5可以在免疫反应过程中将免疫系统中的T/B淋巴细胞和嗜酸性粒细胞等细胞募集到相应区域。此外，它还参与促进结缔组织的粘附和重塑。CXCL5在多种肿瘤组织中高表达。CXCL5可作为肿瘤预后的潜在指标。在肿瘤微环境（TME）中，CXCL5与其受体（例如CXCR2）结合，参与免疫细胞的募集并促进血管生成、肿瘤生长和转移。酸性TME或TNF-α激活的间充质干细胞（MSC）可以分泌CXCL5和其他促炎因子。此外，由纤溶酶原激活物抑制剂-1（PAI-1）产生的癌症相关间皮细胞可以通过NF-κB信号通路分泌IL-8和CXCL5，从而促进腹膜转移。脂肪组织源性干细胞 (ASC) 分泌 CXCL5，随后促进肿瘤生长并影响乳腺肿瘤的发展^[2]。

Recombinant mouse CXCL5 protein (1 μg/50 μL; intravenously injection) injected into WT and Cxcl5^{−/− mice, the results shows that the plasma concentrations of CXCL5, CXCL1, and CXCL2 in Cxcl5−/− mice are attenuated relative to those in WT mice. CXCL5 inhibits chemokine scavenging in blood in part through binding to erythrocyte DARC[4].}

Recombinant mouse CXCL5 (300 ng/mL) blocks insulin-stimulated glucose uptake in mouse embryonic fibroblasts (MEFs). CXCL5 treatment inhibits insulin-induced Akt phosphorylation. CXCL5 blocks insulin signaling by activating the Jak2/STAT5/SOCS2 pathway^[5].

The biological activity determined by a chemotaxis bioassay using human peripheral blood neutrophils is in a concentration of 10-100 ng/ml.
