Toluidine Blue (Toluidine Blue O) is an alkaline quinonimine dye (vivo dye) with high affinity for acidic tissue components, staining nuclei blue and polysaccharides purple. Toluidine Blue shows heterostaining properties for mast cells, mucins and chondrocytes. Toluidine Blue can stain different components of plant tissues and cells in different colours. Toluidine Blue is also used as a diagnostic aid to identify malignant lesions, such as cancer[1][2][3].

体外研究
(In Vitro)

检测浸润性肥大细胞[1]：
1. 组织处理
1) 将组织样本浸入 10% 缓冲福尔马林中 24 小时。
2) 从固定液中取出组织并放入 70% 酒精中。
3) 石蜡包埋：依次在 70%、85%、95%、100% 乙醇、二甲苯、熔融石蜡中孵育（30分钟），并将组织置于石蜡块中。
4) 将石蜡包埋的组织切成 5-7 μm 的切片并置于载玻片上。
5) 使用前将载玻片在 55°C 下烘烤至少 1 小时。
2. 组织染色
1) 将载玻片浸入二甲苯（或 Histo-Clear）5 分钟，对石蜡包埋的组织切片进行脱蜡，然后将组织连续浸入（每次 5 分钟） 100%、95% 和 70% 乙醇，最后 2 分钟浸入蒸馏水中。
2) 将玻片放入 Harris 苏木精溶液中 70 秒。
3) 将载玻片浸入干净的自来水中两到三次，短暂清洗多余的苏木精。在纸巾上轻轻拍打载玻片，除去多余的水。
4) 将载玻片在 70% 酒精中短暂浸泡 3 次，然后用干净的自来水再次清洗 3 分钟。 除去多余的水。
5) 将载玻片放入 Scott 蓝化液中最多一分钟。此步骤可将苏木精转变成细胞核内不溶的蓝色。
6) 在强光显微镜下检查载玻片，确保细胞核正确染色。
7) 用干净的自来水清洗载玻片 3 分钟，并除去多余的水。
8) 将载玻片浸入干净的 ~1.1% Toluidine Blue 溶液中 4 分钟。
(注 1) 9) 用干净的自来水清洗载玻片 3 分钟，并除去多余的水。
10) 将载玻片快速浸入一批 70% 乙醇中 3 次，然后再浸入 5% 伊红溶液中 5 次。
11) 将载玻片连续浸入 70%（2 分钟）、95%（两次，每次 2 分钟）和 100% 乙醇（两次， 每次 2 分钟）； 通过将载玻片浸入二甲苯（或 Histo-Clear）（两次，每次 3 分钟）来结束此步骤。
12) 使用二甲苯封固剂，用干净的盖玻片封固每张载玻片。
13) 让载玻片干燥至少 60 分钟，然后在强光显微镜下进行分析。
注 1：1.1% Toluidine Blue 溶液：将 1.1 g Toluidine Blue 与 100 mL 0.1 M 醋酸钠缓冲液 (pH 4) 混合。搅拌均匀。 通过滴加 1 M 盐酸将 pH 调节至 2.0-2.5。 染色在25℃、避光下进行。
Toluidine Blue 的应用[2][3]：
1.结缔组织粘蛋白，尤其是酸性粘蛋白。 组织被染成紫色到红色，而背景被染成蓝色。
2. 由于肝素和组胺的存在，肥大细胞颗粒呈紫色。
3. 淀粉样蛋白呈蓝色，但在偏振光下呈现亮红色双折射。
4. 内分泌细胞颗粒被染成紫色至红色（染色浓度为 0.01%）。
5. 硫脂呈红棕色或黄色。只有酸性足以引起异染性位移的脂质才会被染色。
6. 白喉棒杆菌含有聚合无机聚磷酸盐颗粒，呈红紫色。
7. 螺杆菌在不同的蓝色背景下染成深蓝色（染色浓度为 1%）。
8. Toluidine Blue 可用于冷冻切片染色，因为染色过程速度快（10-20 s）且细胞清晰度更高。
9、Toluidine Blue 还可将植物的木质素染成蓝绿色，韧皮部染成蓝紫色，植物其余部分染成淡蓝绿色。

  • Do most proteins show cross-species activity?
