Tyrosinase (EC 1.14.18.1) (Polyphenol oxidase) is a rate-limiting enzyme that controls the production of melanin and is encoded by TYR gene. Tyrosinase is mainly found in melanosomes synthesized by skin melanocytes[1].

酶液配制
以 1 mg/mL 的浓度溶于 ddH2O 中。再进一步稀释至浓度为 200-400 U/mL。
实验方法
将分光光度计调整到 280 nm 和 25℃。按照以下顺序将样品加入每个比色管中：
1mL 0.5 M Phosphate buffer, pH 6.5
1 mL 0.001 M L-Tyrosine
0.9 mL ddH2O
通过毛细管将氧气充入比色管中 4-5 分钟，使该反应混合物氧化。将比色管置于分光光度计上，记录 4-5 分钟的 A280，记录为空白组吸光度。加入 0.1 mL 适当稀释的酶，记录 10-12 分钟的 A280。
计算
1U=每分钟引起 280 nm 处吸光度增加0.001的酶量。
