VISTA-IN-4 (Compound B3) is the orally active inhibitor for immune checkpoint protein VISTA with a KD of 0.452 μM. VISTA-IN-4 degrades VISTA in HepG2 with a 77% degradation rate at 5 μM. VISTA-IN-4 activates VISTA-mediated immunosuppression, restores T cell activity, and promotes the release of cytokine IL-2. VISTA-IN-4 exhibits antitumor activity in CT26 xenograft mouse models, and exhibits good pharmacokinetic properties with a good oral exposure in serum with an AUC(0-t) of 412.9 mg·h/mL in SD rats[1].
