Zymosan A (Zymosan A from Saccharomyces cerevisiae) is a TLR2 agonist, that targeting TLRs can prevent and suppress IR-induced intestinal injury. Zymosan A exhibits a significant radioprotective effect, and protects IR-induced intestinal injury in mice. Zymosan A promotes the regeneration of intestinal stem cells (ISCs), after IR injury[1].

体内研究
(In Vivo)

Dimethylnitrosamine 在比格犬中的药代动力学研究显示，静脉注射后的 Dimethylnitrosamine 浓度双相下降，具有 19 分钟的平均分布半衰期和 73 分钟的平均消除半衰期。在低剂量 (0.5和1.0 mg/kg) 下 Dimethylnitrosamine 的消除速率与其血药浓度成正比，但在高剂量 (5.0 mg/kg) 下代谢饱和。Dimethylnitrosamine 的平均全身清除率为 43.3 mL/min/kg，稳态分布体积为 1.9 L/kg，平均停留时间为 45 分钟，且口服生物利用度高达93%。

致病原理
Zymosan A 可以介导包括IL-1β、TNF-α、HMGB1 等促炎细胞因子的释放，诱导炎症反应，导致腹膜炎。
具体造模方法
小鼠: C57BL/6 • 雄性 • 3-7 月龄
给药方式: 100 或 500 mg/kg • 腹腔注射
Note
(1) 将 ChAT-eGFP 小鼠随机分为两组，注射前测量每组的基线直肠温度和心电图 (ECG)。
(2) Zymosan A 悬浮液的制备: 称量 Zymosan A 并用紫外线消毒 30 分钟，加入无菌过滤盐水并充分涡旋，室温保存过夜，并在使用前再次涡旋以确保充分混合。
(3) 分别在 Zymosan A 注射 4 小时和 24 小时后每小时记录直肠温度和心电图。
造模成功指标
表型观察: 体温过低；心功能不全，包括基线心率 (HR) 变异性增加，心室传导适度减慢，心脏收缩力降低；细菌转移到腹膜腔和血液中。
