生物活性

BODIPY-FL is a green fluorescent dye that can be used to label probes or primers. BODIPY-FL fluorescence can be quenched after interacting with uniquely positioned guanine, making it useful for quantifying specific DNA or RNA molecules. BODIPY-FL-labeled sphingolipid analogs can be used to investigate sphingolipid internalization, transport, and endocytosis in mouse embryonic stem cells. BODIPY-FL-labeled monoterpenes can quickly penetrate bacteria, mammalian, and fungal cells, allowing for the detection of characteristics of a broad spectrum of Gram-positive and Gram-negative bacteria, as well as pathogenic fungi. The maximum absorption wavelength of BODIPY-FL is 505 nm, and the maximum emission wavelength is 513 nm[1][2][3][4].

BODIPY FL-鞘磷脂染色液的配制[3]
1.1 配制储存液
用氯仿/乙醇溶剂 (19: 1 (v/v)) 配制 1 mM 的 BODIPY FL-鞘磷脂原液。
注:1) BODIPY FL-鞘磷脂储存液转移至琥珀色玻璃圆形样品瓶中并于 -20 ℃ 避光保存。
1.2 配制工作液
1) 将氮气蒸发器的喷嘴放入琥珀色玻璃圆形小瓶中,并将氮气喷向储存液,直至液相完全干燥;再加入 200 µL 无水乙醇,以重构干燥的 BODIPY FL-鞘磷脂粉末。
2) 制备 5 µM 脱脂 BSA 溶液,将适量脱脂 BSA 溶解在 10 mL 含有 10 mM HEPES (HBSS/HEPES 缓冲液) 的 Hanks 缓冲盐溶液中。
3) 将 1) 中制备的 200 μL BODIPY FL-鞘磷脂乙醇溶液加入 2) 中制备的 10 mL 脱脂 BSA 溶液中,在涡流混合器上混合,得到工作液 (5 μM BODIPY FL-鞘磷脂 + 5 μM BSA)。
注:1) 工作液于 -20 ℃ 避光保存。
BODIPY FL-鞘磷脂染色
1) 用 ESGRO 生长培养基培养 mESC 细胞,并将细胞以 5 × 104 个细胞/cm2 的密度接种于培养皿中,37 ℃、5% CO2 培养箱培养过夜。
2) 将过夜细胞培养皿中的生长培养基吸出,并用 HBSS/HEPES 缓冲液洗涤贴壁细胞 3 次。
3) 将 BODIPY FL-鞘磷脂工作液加入培养皿中,完全覆盖细胞生长区域,并在 4 ℃ 下避光孵育细胞 30 分钟。
4) 去除所有工作液,并用 ESGRO 生长培养基洗涤细胞 3 次。
5) 加入 RT ESGRO 生长培养基以覆盖细胞,并在 37 ℃、 5% CO2 的培养箱中避光孵育细胞 30 分钟。
6) 用 ESGRO 生长培养基清洗细胞,用 LSM 780 紫外共聚焦显微镜对染色细胞进行检查和成像。
7) 数据分析 (共聚焦显微镜):含有高水平 BODIPY FL-鞘脂类似物的细胞器或区域发出红色荧光 (λem ≥ 590 nm),含有低水平 BODIPY FL-鞘脂类似物的细胞器或区域发出绿色荧光 (λem = 520-590 nm)。
