Mirikizumab  (Synonyms: 米吉珠单抗; LY­3074828)

Mirikizumab (LY3074828) is a humanized IgG4 monoclonal antibody that targets the p19 subunit of interleukin 23 (IL-23). Mirikizumab binds to human and monkey IL-23 with high affinity, with K_d values of 21 pM and 55 pM, respectively. By inhibiting the binding of IL-23 to IL-23R, Mirikizumab modulates the immune response and holds potential for research in ulcerative colitis and Crohn's disease^[1][2].

Mirikizumab (4.4 pM - 800 nM，48 小时) 可在小鼠脾细胞中完全中和由人和食蟹猴 IL-23 诱导产生 IL-17A，对人和食蟹猴 IL23 的 IC₅₀ 分别是 82 pM 和 120 pM^[2]。
Mirikizumab (0.1 ng/mL - 1 μg/mL, 48 小时) 能够在两组人外周血单核细胞 (PBMCs) 中完全抑制由抗 CD3 /抗 CD28 及 IL-23 诱导的 IL-17 分泌，对这两组 PBMCs 的IC₅₀ 分别是 22 μg/mL 和 29 μg/mL^[2]。
Mirikizumab (50 or 500 ng/mL，48 小时) 不会影响 Kit225 和 TALL-104 细胞中 IL-12 诱导的 STAT4 磷酸化^[2]。
