Reverse Phase Chromatography is used to separate mixtures of molecules with different hydrophobic and hydrophilic properties. The technique uses silica gel matrices with covalently bound n-alkyl chains as the stationary phase. For example, C-18 columns strongly retain hydrophobic compounds, while hydrophilic molecules elute more quickly. This method is widely used in the separation and purification of proteins, peptides, and small molecule drugs.
