Neutral protease I  (Synonyms: Dispase I)

Neutral protease I (Dispase I) is a rapid, effective, gentle and neutral protease that can separate intact epidermis from the dermis. Neutral protease I can also separate intact epithelial sheets in culture from the substratum. Neutral protease I preserves the viability of the epithelial cells while cleaving the basement membrane zone region. Neutral protease I can also be used to prevent clumping in suspension cultures. Neutral protease I cleaves fibronectin and type IV collagen, but not laminin, type V collagen, serum albumin, or transferrin^[1].

使用方法
1、溶液配制
1) 用 DPBS 缓冲盐溶液 (无钙镁离子) 溶解适量本品冻干粉，配制成 10 mg/mL 的储存液，用 0.22 μM 滤膜过滤除菌。
2) 使用时用 DPBS 将上述储存液稀释到工作液浓度即可，用于细胞分离时其常用工作浓度为 0.6-2.4 U/mL。
注：不推荐使用高于 2.4 U/mL 的工作浓度。
2、组织的解离
1) 用无菌的小刀或者剪刀将组织剪成 3-4 mm 的组织块；
2) 用无菌 PBS 清洗组织块；
3) 向组织块中加入 Dispase II 溶液 (工作浓度为 0.6-2.4 U/mL)，并确保组织块全部浸没于 Dispase 溶液中。
4) 37℃ 孵育，孵育过程缓慢搅拌直至组织块全部解离。
注：一般对于较难解离组织，1h 即可达到分离目的，但更长时间 (如数小时) 的孵育也不会明显影响细胞活性。
5) 如有需要，可将上述消化产物通过无菌不锈钢网筛过滤，以分开单细胞与残留的组织块。或者待大块组织沉淀后轻轻倒出上层细胞，若是需要，换用新鲜 dispase 溶液以进一步解离残留组织。
6) 离心沉淀细胞，倒掉酶溶液；
7) 用培养基重悬细胞沉淀，并于常规条件下培养细胞。
3、细胞传代
1) 利用 Dispase 溶液 (37℃预热) 浸没细胞，于 37℃ 孵育 5 min；
2) 吸除上述溶液，继续于 37℃ 孵育 10 min；
3) 显微镜下观察细胞分离情况，如需要，可进一步孵育 15 min；
4) 利用细胞培养基悬浮细胞，轻轻旋转使得细胞沉淀用培养基清洗细胞；
5) 换用新鲜细胞培养液重悬细胞，并按常规方法进行细胞铺板。
