生物活性

Physalin A is a biologically active withanolide. Physalin A has been shown to be anti-inflammatory, antifibrotic and ameliorative of autophagy in models of disc degeneration. Physalin A has antitumor activity in non-small cell lung cancer cells, inducing apoptosis, ROS production and G2/M phase cell cycle arrest. Besides. Physalin A can significantly increase the activity of quinone reductase and increase the expression of detoxifying enzymesc[1][2][3][4].

(In Vitro)

Result: Increased the phosphorylation levels P38 and ERK in a time-dependent manner.

Cell Line: NP cells were harvested from the inner tissue of tail disc of SpragueDawley rats

Incubation Time: 48 h
Result: Reduced the phosphorylation levels of IKKα/β, IκBα and P65.
Reduced the phosphorylation levels of JNK, ERK and P38.
Reduced the phosphorylation levels of PI3K. AKT and mTOR.

Western Blot Analysis[4]

Cell Line: H292, H358 and H1975 cells
Concentration: 0, 2.5, 5, 10 and 15 μM
Incubation Time: 24 h
Result: Increased the levels of apoptotic markers, such as cleaved poly(ADP-ribose) polymerase (PARP), cleaved caspase-3 and cleaved caspase-9.

Physalin A (2 μl 20 μM；椎间盘髓核内注射；单剂量) 在椎间盘退变大鼠模型中具有保护作用[3]。
Physalin A (40-80 mg/kg；腹腔注射；10 天) 在 H292 异种移植瘤模型中具有抗肿瘤的作用[4]。

Animal Model: Male Sprague-Dawley rats aged 6-8 weeks old (220±20 g) with intervertebral disc degeneration[3]
Dosage: 2 μl 20 μM
Administration: Injection of nucleus pulposus of the disc; Single dose
Result: Alleviated disc degradation.
Reduced inflammation and fibrosis.
Animal Model: Male BALB/c mice[4]
Dosage: 40 and 80 mg/kg
Administration: Intraperitoneal injection (i.p.); 10 days
Result: Significantly reduced the mean tumor volume and tumor weight, had the least effect on the weight of mice.
Decreased Tyr705-p-STAT3 levels, and increased caspase-3 activation.
