生物活性

SA91-0178 is a METTL1 inhibitor. SA91-0178 inhibits m7G methylation of RNA, reduces SARM1 stability, mitigates NAD+ depletion and metabolic reprogramming in macrophages. SA91-0178 demonstrates excellent protective efficacy against multiple organ injury in cecal ligation and puncture (CLP)-induced and ischemia/reperfusion (I/R)-induced mice. SA91-0178 can be used for the study of systemic inflammatory diseases[1].

Animal Model: 8-week-old male C57BL/6 mice, Lyz2creMettl1fl/fl mice were ligated and punctured, and a small amount of cecal content was extruded by gentle squeezing [1]
Dosage: 5, 10, 15 mg/kg
Administration: ip, once
Result: Achieved reduction rates of serum Cre, BUN, AST, and ALT.
Reduced the renal tubular injury score.
Appeared decreased F4/80+ macrophage infiltration in kidney, liver, lung, and heart.
Showed greater reduction in SARM1 mRNA/protein levels in F4/80+ macrophages.
