生物活性

Aflatoxin B2 is an orally active hepatotoxic mycotoxin and an inducer of apoptosis/autophagy in hepatocytes. Aflatoxin B2 triggers mitochondria-mediated apoptosis by increasing ROS generation, promoting the translocation of Bax and cytochrome C, activating caspase-3 and PARP cleavage. Aflatoxin B2 also induces autophagy by inhibiting the PI3K/Akt/mTOR signaling pathway, upregulating Beclin-1 and LC3-II/LC3-I, and downregulating p62. Aflatoxin B2 can be used to construct a liver injury model[1][2][3].

Cell Viability Assay[1]

Cell Line: J774A.1 murine macrophages
Concentration: 10 μM, 50 μM, 100 μM
Incubation Time: 24 h and 48 h
Result: MTT assay showed no significant cytotoxicity at 10-50 μM, while 100 μM Aflatoxin B2 significantly reduced cell viability by 30% at 48 h, indicating dose-dependent cytotoxicity at high concentrations.
Flow cytometry with Annexin V/PI staining revealed no significant increase in apoptotic cells at 10-100 μM, suggesting cytotoxicity was not mediated by classical apoptotic pathways.
Western blot analysis showed no significant changes in caspase-3 or PARP cleavage at 10-50 μM, consistent with the lack of apoptotic induction.

Animal Model: Cobb500 broilers (1-day-old, 40-50 g)[3]
Dosage: 0.2 mg/kg, 0.4 mg/kg, 0.8 mg/kg (add in feed)
Administration: Oral administration via feed, once daily, for 21 consecutive days
Result: Dose-dependent increased in apoptotic hepatocytes with nuclear fragmentation and chromatin condensation. Revealed crescent-shaped nuclei and mitochondrial swelling.
Activated cleaved caspase-3 and PARP, indicating activation of the mitochondrial apoptotic pathway. Intracellular reactive oxygen species (ROS) levels increased significantly, promoting Bax translocation to mitochondria and cytochrome c release into the cytosol, forming apoptosomes with Apaf-1 and caspase-9.
Enhanced MDC staining and LC3 immunofluorescence formation of autophagic vacuoles and LC3 puncta. Upregulated Beclin-1, increased LC3-II/LC3-I ratio, and downregulated p62, confirming autophagic flux.
Inhibited the PI3K/Akt/mTOR signaling pathway, as evidenced by reduced phosphorylation of PI3K, Akt, and mTOR (p-PI3K, p-Akt, p-mTOR S2448) and upregulated PTEN expression.
Upregulated Bax mRNA and downregulated Bcl-2 mRNA, while Western blot of mitochondrial and cytosolic fractions revealed increased Bax in mitochondria and cytochrome c in the cytosol, supporting mitochondrial pathway activation.
