在对中国药用植物的新农药筛选项目中,发现 Notopterygium incisum 根茎的乙醇提取物对两种线虫 Bursaphelenchus xylophilusMeloidogyne incognita 具有强烈的杀虫活性。根据生物活性指导的分级分离,从乙醇提取物中分离出四种成分,并鉴定为 Columbianetin、Falcarindiol、Falcarinol 和 Isoimperatorin。Isoimperatorin 对 B. xylophilus 的 LC50 值为 21.83 μg/mL。当使用 15 分钟的紫外光处理时,falcarindiol、falcarinol 和 isoimperatorin 对南方根结线虫的毒性比在黑暗处理时几乎增加了五倍,而 columbianetin 仅显示出两倍的毒性。 Isoimperatorin 被证明对多种昆虫(如甘蓝蚜 (Brevicoryne brassicae ))具有杀虫活性[2]。Isoimperatorin 在 Angelica dahurica (AD) 提取物的活性分级中被鉴定出来[3]。Isoimperatorin 通常作为内标 (IS) 使用[4]

  • 方案 一

    请依序添加每种溶剂: 10% EtOH    40% PEG300    5% Tween-80    45% Saline

    Solubility: ≥ 0.83 mg/mL (3.07 mM); 澄清溶液

    此方案可获得 ≥ 0.83 mg/mL(饱和度未知)的澄清溶液。

    1 mL 工作液为例,取 100 μL 8.3 mg/mL 的澄清 EtOH 储备液加到 400 μL PEG300 中,混合均匀;再向上述体系中加入 50 μL Tween-80,混合均匀;然后再继续加入 450 μL 生理盐水 定容至 1 mL

    生理盐水的配制:将 0.9 g 氯化钠,溶解于 ddH₂O 并定容至 100 mL,可以得到澄清透明的生理盐水溶液。

    请依序添加每种溶剂: 10% EtOH    90% Corn Oil

    Solubility: ≥ 0.83 mg/mL (3.07 mM); 澄清溶液

    此方案可获得 ≥ 0.83 mg/mL(饱和度未知)的澄清溶液,此方案实验周期在半个月以上的动物实验酌情使用。

    1 mL 工作液为例,取 100 μL 8.3 mg/mL 的澄清 EtOH 储备液加到 900 μL 玉米油中,混合均匀。
